Enhanced Antitumor Efficacy through an “AND gate” Reactive Oxygen?Species?Dependent pH?Responsive Nanomedicine Approach

Anticancer drug delivery strategies are designed to take advantage of the differential chemical environment in solid tumors independently, or high levels reactive oxygen species (ROS) low pH, compared healthy tissue. Here, design and thorough characterization two functionalizable “AND gate” multiresponsive (MR) block amphiphilic copolymers reported, aimed full coexistence cues—ROS pH—present tumor microenvironment. The hydrophobic blocks contain masked pH-responsive side chains, which exposed exclusively response ROS. Hence, polymer chains will undergo a charge shift very relevant pH window present extracellular milieu most (pH 5.6–7.2) after demasking by Doxorubicin (DOX)-loaded nanosized MR polymersomes (MRPs) fabricated via microfluidic self-assembly. Chemical reveals ROS-dependent sensitivity accelerated DOX release under influence both ROS pH. Treatment tumor-bearing mice with DOX-loaded nonresponsive MRPs dramatically decreases cardiac toxicity. optimal outperform free terms growth inhibition survival, shedding light on requirements for successful cancer nanomedicine.
